This preprint studies sleep problems in Kleefstra syndrome caused by EHMT1 variants. It reports severe sleep-maintenance insomnia in many people with KLEFS1 and uses fruit fly models to link EHMT1/G9a to fragmented sleep, metabolism, oxidative stress, and insulin-producing cells. Developmental antioxidant treatment and a sleep-restriction paradigm restored sleep continuity in flies.
