A new study sheds light on developmental regression in Kleefstra syndrome, identifying astrocytes and the inflammatory protein S100B as key contributors. In an international cohort, many individuals treated with olanzapine showed improved adaptive functioning. Laboratory studies suggest olanzapine reduces astrocyte-driven neuronal hyperactivity, highlighting a promising therapeutic target for future treatments.
